Modeling of the Technological Process for Live Rotavirus Vaccine Production

We simulate the industrial technology for rotavirus vaccine, based on the strain of group A rotavirus with G1P[8] genotype, circulating among people in Ukraine. We propose to use this strain of rotavirus as the industrial strain. Through successive passages, it is adapted to cultivation conditions in the continuous human and animal cell cultures. The model of technology for the rotavirus vaccine, based on the industrial strain of rotavirus, is developed for its cultivation in continuous cell culture Vero.

Adaptation to cultivation conditions in vitro and increase of infectious titres of human rotaviruses, isolated on the territory of Ukraine

In this paper, we isolate and adapt five strains of rotaviruses for cultivation conditions from children with acute intestinal infections in HEP-2 human cell culture. We characterize the isolated strains ding to genotype and ability to increase the infectious titres during five consecutive passages. Crucially, we isolate the rotavirus strain, which has a special ability to adapt to cultivation conditions in human cell culture. This strain is promising for creating effective rotavirus vaccine for Ukrainians, as well as for developing diagnostic test-systems.

The features of human rotaviral selective biotechnology in cell culture and their adaptation to cultivation conditions

In the recent years rotaviral infection has proliferated in Ukraine. The problem is that the scientists have not yet determined which human strains of rotaviruses circulate in its territory and their biological properties. Hence we develop the biotechnology of human rotaviruses isolation in the cell culture and their adaptation to the cultivation conditions. We also study their biological properties and create the collection of rotavirus strains, circulating in the territory of the Ukraine.
